Biological Background: CD11c Function and Localization
- CD11c, also known as Integrin alpha-X, is a member of the integrin family and the CD11 antigen-like family. It is encoded by the Itgax gene in mice.
- Integrin alpha-X pairs with integrin beta-2 to form the leukocyte adhesion receptor p150,95. This heterodimer serves as a receptor for fibrinogen, recognizing the G-P-R sequence.
- The p150,95 integrin mediates cell-cell interactions during inflammatory responses, playing a crucial role in monocyte adhesion and chemotaxis.
- Additionally, Integrin alpha-X functions as a receptor for the erythrocyte-specific adhesion molecule ICAM4, facilitating erythrophagocytosis.
- CD11c is predominantly localized to the cell membrane as a single-pass type I transmembrane protein.
- Consistent with its role in adhesion and signaling, Integrin alpha-X is a glycoprotein containing disulfide bonds and calcium/magnesium-binding domains, characteristic of integrin receptors.
Experimental Guidance and Technical Tips
- This antibody is directly conjugated to PE/Cyanine7, making it suitable for use in flow cytometry (FC) without the need for a secondary antibody.
- Based on the recombinant protein immunogen, the antibody may recognize both native and denatured forms of CD11c, but validation in the relevant sample system is recommended.
- Titration experiments should be considered to determine the optimal antibody concentration for specific cell types and flow cytometry protocols.
- Since CD11c is expressed on leukocytes, particularly myeloid cells, use appropriate mouse leukocyte samples (e.g., spleen, blood, bone marrow) for testing.
