  • CD45, also known as leukocyte common antigen (L-CA), lymphocyte antigen 5 (Ly-5), and T200, is a receptor-type tyrosine-protein phosphatase C encoded by the Ptprc gene. Multiple isoforms are generated via alternative splicing, including CD45R and B220, with molecular weights ranging from 130 to 144 kDa.
  • It functions as a critical regulator of T-cell activation through the antigen receptor, acting as a positive coactivator upon binding to DPP4. The first of its two intracellular phosphatase domains has enzymatic activity, while the second modulates substrate specificity.
  • CD45 dephosphorylates key signaling molecules such as SKAP1, FYN, and LYN to modulate their activity. It also interacts with CLEC10A on dendritic cells, linking glycosylation sensing to T-cell activation thresholds.
  • The protein localizes to the cell membrane, membrane rafts, and immunological synapse, positioning it as a key mediator of signaling at T-cell–antigen-presenting cell contacts.
  • CD45 is heavily modified by glycosylation and phosphorylation, as reflected by its UniProt keywords: glycoprotein, phosphoprotein. It spans the membrane via a single transmembrane helix.
