  • CD45R/B220, encoded by the Ptprc gene, is a receptor-type tyrosine-protein phosphatase (PTPase) also known as Leukocyte Common Antigen (L-CA) and Lymphocyte Antigen 5 (Ly-5). It is predominantly expressed on hematopoietic cells, including B and T lymphocytes, and exists in multiple isoforms generated by alternative splicing.
  • The protein serves as a critical regulator of T-cell receptor signaling by modulating the activity of Src family kinases such as LYN and FYN. It dephosphorylates LYN, influencing downstream signaling cascades essential for T-cell activation and coactivation upon binding to DPP4.
  • At antigen-presenting cell (APC)–T cell immunological synapses, CD45 interacts with CLEC10A on immature dendritic cells, which recognizes Tn antigen-carrying CD45 on effector T cells. This interaction helps fine-tune T-cell activation thresholds and limit autoreactive responses (By similarity).
