Biological Background: IFN-gamma Function and Localization
- Interferon gamma (IFN-gamma) is a type II interferon cytokine mainly produced by activated T lymphocytes and natural killer (NK) cells, acting as a central mediator of innate and adaptive immunity.
- It exerts antimicrobial, antiviral, and antitumor activities by activating effector immune cells and enhancing antigen presentation via both MHC class I and II pathways.
- Signals through the JAK-STAT pathway: upon binding to its receptor IFNGR1, it induces activation of JAK2, JAK1, and STAT1, leading to STAT1 nuclear translocation and transcription of interferon-regulated genes such as IRF1.
- Upregulates MHC class I antigen processing by inducing replacement of constitutive proteasome subunits with immunoproteasome subunits and by inducing the PA28 activator, thereby enhancing peptide generation and repertoire for MHC loading.
- Promotes MHC class II expression by upregulating key molecules such as cathepsins B, H, and L (By similarity).
- Participates in hematopoietic stem cell regulation, affecting development, quiescence, and differentiation (By similarity).
- The protein is secreted and contains characteristic structural features, including a signal peptide, glycosylation sites, and a pyrrolidone carboxylic acid modification.
