Biological Background: CD54 Function and Localization

• ICAM-1 (CD54) is a cell adhesion molecule that serves as a ligand for the integrin receptors LFA-1 (ITGAL:ITGB2) and MAC-1 (ITGAM:ITGB2), mediating leukocyte adhesion and immune synapse formation.
• It plays a critical role in leukocyte trans-endothelial migration by promoting the assembly of endothelial apical cups through ARHGEF26/SGEF and RHOG activation.
• ICAM-1 facilitates cell aggregation in epithelial cells via interaction with MUC1.
• The protein is a single-pass type I membrane glycoprotein containing immunoglobulin-like domains, and undergoes alternative splicing, disulfide bond formation, glycosylation, and ubiquitin conjugation.
• Subcellularly, ICAM-1 localizes to the cell membrane.
• Expression: low basal level on subsets of lymphocytes, macrophages, and endothelial cells, but strongly induced upon stimulation on these and on fibroblasts and epithelial cells.
