Biological Background: CD27 Function and Localization
- CD27 (T-cell activation antigen CD27, TNFRSF7) is a single-pass type I transmembrane receptor belonging to the tumor necrosis factor receptor superfamily.
- It serves as a costimulatory immune-checkpoint receptor on T-cells, NK-cells, and B-cells; binding to its ligand CD70/CD27L on B-cells triggers the CD70-CD27 signaling pathway, mediating antigen-specific T-cell activation and expansion for B-cell immune surveillance. - CD70 ligation activates the TRAF2-PTPN6 axis, inhibiting LCK phosphorylation to promote phenotypic and transcriptional adaptations of T-cell memory (By similarity).
