For optimal staining, perform fixation and permeabilization appropriate for nuclear/cytoplasmic proteins and titrate the PE-conjugated antibody to determine the optimal signal-to-noise ratio.
  • S100A9 is highly expressed in myeloid lineage cells (neutrophils, monocytes, macrophages). Use relevant positive controls such as mouse bone marrow or splenocytes, and include isotype controls and Fc blocking to minimize non-specific binding.
