Biological Background: LAMP1/CD107a Function and Localization

  • LAMP1 (Lysosome-associated membrane glycoprotein 1), also known as CD107a, LGP-120, LGP-A, P2B, and Lamp-1, is a heavily glycosylated transmembrane protein encoded by the Lamp1 gene in mice.
  • It plays a critical role in lysosome biogenesis, pH regulation, autophagy, and cholesterol homeostasis by acting as a direct inhibitor of the proton channel TMEM175, thereby facilitating lysosomal acidification (PubMed:15121881). Related references: PMID:15121881
  • In natural killer (NK) cells, LAMP1 is essential for cytotoxicity, mediating cytotoxic granule movement and perforin trafficking to the lytic granule, while also protecting NK cells from self-degranulation damage (PubMed:23847195). Related references: PMID:23847195
  • It localizes to the membranes of lysosomes, late endosomes, and cytolytic granules, and can also be present on the cell surface upon degranulation.
