Biological Background: IL-9 Function and Localization
- Interleukin-9 (IL-9), also known as cytokine P40 and T-cell growth factor P40, is a multifunctional cytokine encoded by the Il9 gene in mouse (UniProt P15247).
- IL-9 is primarily secreted by T-helper 2 (Th2) lymphocytes, mast cells, and NKT cells, and plays critical roles in immune responses against parasites, as well as in allergic inflammation. Related references: PMID:11070175, PMID:19433802
- It modulates intestinal epithelial permeability and contributes to adaptive immunity, particularly in mucosal tissues. Related references: PMID:12704113

- The immunogen corresponds to amino acids 19–114 of mouse IL-9 (NP_032399.1), which encompasses the mature secreted protein lacking the signal peptide. This design is likely to generate antibodies recognizing the native, processed form of IL-9.
- As a PE-conjugated monoclonal antibody, it is optimized for intracellular flow cytometry (FC intra). For detection of intracellular IL-9, cells should be fixed and permeabilized using a suitable protocol prior to staining. A stimulation step (e.g., with PMA/ionomycin in the presence of a protein transport inhibitor) may be required to accumulate cytokine intracellularly in certain cell types.
- Since the antibody is reported to react with mouse IL-9, cross-reactivity with human or other species has not been evaluated; it is recommended to validate specificity in the intended sample system.
- The monoclonal rabbit origin may offer high affinity and specificity; however, titration is advisable to determine the optimal concentration for each experimental condition.
