Biological Background: IL-10RA/CD210 Function and Localization
- Interleukin-10 receptor subunit alpha (IL-10RA, CDw210a, IL-10R1) is a cell surface receptor for the cytokine IL-10, playing a key role in limiting excessive tissue disruption caused by inflammation.
- Upon IL-10 binding, IL-10RA recruits IL-10RB to form a heterotetrameric complex, which activates associated JAK1 and TYK2 kinases, leading to phosphorylation of specific tyrosine residues on IL-10RA and subsequent STAT3 recruitment and phosphorylation (PubMed:8910398). - Phosphorylated STAT3 homodimerizes and translocates to the nucleus to induce anti-inflammatory gene expression.
- IL-10RA-mediated activation of STAT3 also inhibits starvation-induced autophagy (By similarity).
