Biological Background: E-Cadherin/CD324 Function and Localization

  • E-Cadherin/CD324 is a calcium-dependent cell adhesion protein that preferentially interacts homophilically to connect cells, contributing to sorting of heterogeneous cell types and regulating cell-cell adhesions, mobility, and proliferation of epithelial cells. Related references: PMID:11976333
  • It promotes organization of radial actin fiber structure and cellular response to contractile forces via interaction with AMOTL2, and facilitates desmosome formation by recruiting DSG2 and DSP.
  • The cleaved fragment E-Cad/CTF2 promotes non-amyloidogenic degradation of Abeta precursors, inhibiting APP C99 and C83 production.
  • Does not function as a receptor for L. monocytogenes internalin A (InlA); a single surface-exposed residue mutation can confer receptor activity.
  • Subcellularly localized to adherens junctions, cell membrane, endosomes, trans-Golgi network, cytoplasm, and desmosomes.
  • Expressed in non-neural epithelial tissues, including keratinocytes, organ of Corti cells, and granuloma macrophages.
  • Post-translational modifications include disulfide bonds, glycosylation, phosphorylation, and ubiquitination.
