Biological Background: C5a anaphylatoxin chemotactic receptor 1 Function and Localization
-
The protein is the C5a anaphylatoxin chemotactic receptor 1 (C5aR, CD88), a receptor for the chemotactic and inflammatory peptide anaphylatoxin C5a. Upon binding, it stimulates chemotaxis, granule enzyme release, intracellular calcium release, and superoxide anion production.

Ligand binding causes a conformational change that triggers signaling via guanine nucleotide-binding proteins (G proteins), modulating the activity of downstream effectors such as adenylate cyclase.
Related references: PMID:37852260 -
The receptor is coupled to G(i)/G(o) G alpha proteins (GNAI1 or GNAO1) and mediates inhibition of adenylate cyclase.

The ligand interacts with at least two sites on the receptor: a high-affinity site on the extracellular N-terminus and a second site in the transmembrane region that activates downstream signaling.
-
Subcellularly, the receptor localizes to the cell membrane and cytoplasmic vesicles. It belongs to the GPCR family and is classified under keywords such as chemotaxis, disulfide bond, glycoprotein, phosphoprotein, and sulfation.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 1-37 of mouse CD88/C5aR, which covers the N-terminal extracellular region. This suggests the antibody likely targets an epitope in the extracellular domain.
- The antibody is PE-conjugated, enabling direct detection in flow cytometry (FC) without a secondary antibody. Consider using appropriate compensation controls for PE fluorescence.
- For FC experiments, validate the antibody in your specific sample system. Include relevant isotype controls and assess optimal staining concentrations.
- Since the antibody is raised in rabbit and reacts with mouse, avoid cross-reactivity with other species unless specified.
