Biological Background: T-lymphocyte activation antigen CD86 Function and Localization
- CD86 (also known as B7-2, ETC-1, Ly-58, and CD28 ligand 2) is a member of the immunoglobulin superfamily and a key costimulatory molecule.
- Functions as a ligand for the CD28 receptor, providing an essential secondary signal that augments MHC/TCR-mediated activation of naive T-cells.
- Acts as an inhibitor of T-cell activation by binding to CTLA4, a decoy receptor that blocks CD28-mediated T-cell priming. Related references: PMID:7504059, PMID:7513726
- Regulates B-cell function by controlling IgG1 production and activating the NF-kappa-B pathway upon CD40 engagement. Related references: PMID:23241883
- Localizes to the cell membrane and is expressed on the surface of antigen-presenting cells.
- Post-translational modifications include glycosylation and disulfide bond formation; also undergoes ubiquitination.
- Alternative splicing produces multiple isoforms, contributing to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 24–245), ensuring recognition of the native conformation for flow cytometry.
- For flow cytometry, titrate the PE-conjugated antibody to determine the optimal staining concentration and include an Fc-blocking step to reduce non-specific binding.
- Use appropriate viability dyes and gates to exclude dead cells during acquisition.
- Validate staining in the relevant sample system (e.g., mouse splenocytes or antigen-presenting cell lines) and include isotype controls.
