Biological Background: SLAM family member 5 Function and Localization
- Encodes SLAM family member 5 (SLAMF5), also known as CD84, CDw84, or Leukocyte differentiation antigen CD84 — a type I transmembrane receptor of the immunoglobulin superfamily with two extracellular Ig-like domains and cytoplasmic ITSMs.
- Functions as a self-ligand receptor, mediating homophilic interactions that modulate both innate and adaptive immune cell activation and differentiation; signaling is dependent on adapter proteins SH2D1A/SAP and SH2D1B/EAT-2. Related references: PMID:20962259
- Promotes natural killer (NK) cell cytotoxicity in a SH2D1A- and SH2D1B-dependent manner. Related references: PMID:20962259
- Required for prolonged T-cell:B-cell contact, optimal T follicular helper function, and germinal center formation. Related references: PMID:20153220
- Maintains B cell tolerance and prevents autoimmunity within germinal centers. Related references: PMID:25801429
- In macrophages, enhances LPS-induced MAPK phosphorylation and NF-κB activation and modulates cytokine secretion via ITSM 2. Related references: PMID:20628063
- Localizes to the cell membrane; predominantly expressed in hematopoietic tissues (lymph node, spleen, thymus, bone marrow) and lung.
- Annotated with keywords including adaptive immunity, autophagy, cell adhesion, disulfide bond, glycoprotein, immunoglobulin domain, innate immunity, phosphoprotein, and alternative splicing.
Experimental Guidance and Technical Tips
- This PE-conjugated antibody is designed for direct flow cytometry (FC) detection of mouse CD84 on live cells; no secondary antibody is required.
- The immunogen encompasses amino acids 22–221 of the extracellular domain, expected to recognize the native receptor; consider titrating (e.g., 0.1–1 µg per 10^6 cells) to determine optimal staining concentration.
- For other applications such as immunofluorescence or immunohistochemistry, validate with appropriate sample preparation, as the PE fluorophore is large and may affect tissue penetration.
- When studying homophilic CD84 interactions, note that the antibody might compete with ligand binding; pre-incubation experiments may help dissect functional effects.
