Biological Background: CD82 Function and Localization

  • CD82, also known as Kai1, Tspan27, IA4, C33 antigen, and inducible membrane protein R2, is a member of the tetraspanin superfamily. It is a structural component of tetraspanin-enriched microdomains (TERMs), which serve as platforms for receptor clustering and signaling.
  • Core functions include attenuation of EGF signaling by facilitating ligand-induced endocytosis and desensitization of EGFR, mediated through ubiquitination and trafficking of EGFR via PKC-dependent CBL phosphorylation. It also modulates cell adhesion and migration by regulating integrins such as alpha4/ITA4, alpha6/ITGA6, and beta1/ITGB1, and interferes with urokinase-type plasminogen activator (PLAU) and its receptor (PLAUR) to decrease plasminogen activation.
