Biological Background: CD62L (L-selectin) Function and Localization
- CD62L, also known as L-selectin, is a calcium-dependent lectin that mediates cell adhesion by binding to glycoproteins on neighboring cells. It plays a key role in lymphocyte homing to peripheral lymph nodes and leukocyte tethering/rolling on endothelium. Related references: PMID:1693096
- The protein is expressed on the surface of most leukocytes and is predominantly found in lymphoid tissues.
- L-selectin is a transmembrane protein with an extracellular region containing a C-type lectin domain, an EGF-like domain, and multiple sushi (SCR) repeats. The cytoplasmic tail is involved in signaling.
- It is rapidly shed from the cell surface upon leukocyte activation, a process mediated by proteolytic cleavage.
- CD62L belongs to the selectin family of adhesion molecules, which also includes E-selectin and P-selectin.
- Post-translational modifications include glycosylation and disulfide bond formation essential for ligand binding.
Experimental Guidance and Technical Tips
- This antibody is raised against the extracellular domain (aa 39-332) and is PE-conjugated, making it suitable for direct flow cytometric analysis of surface CD62L expression on live mouse cells.
- For optimal staining, consider titrating the antibody to determine the best concentration for your specific cell type and flow cytometry conditions.
- Use an appropriate isotype control and Fc receptor blocking to minimize non-specific binding.
- Note that PE is a bright fluorophore; avoid prolonged light exposure to prevent photobleaching.
