Biological Background: CD6 Function and Localization
- CD6 is a T-cell differentiation antigen and a type I transmembrane glycoprotein belonging to the scavenger receptor cysteine-rich (SRCR) superfamily.
- It mediates cell-cell contacts by binding to ALCAM/CD166, functions as a costimulatory molecule that promotes T-cell activation and proliferation, and contributes to the formation and maturation of the immunological synapse.
- Contributes to signaling cascades triggered by activation of the TCR/CD3 complex. Related references: PMID:24584089
- Functions as a calcium-dependent pattern recognition receptor that binds both Gram-positive and Gram-negative bacteria via lipoteichoic acid and lipopolysaccharide (LPS). LPS binding activates downstream MAP kinases, leading to pro-inflammatory cytokine secretion.
- Localized to the cell membrane.
- Predominantly expressed in thymus, lymph node, and spleen.
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation.
