Biological Background: CD5 Function and Localization
-
CD5 is a lymphoid-specific receptor expressed on all T-cells and a subset of B-cells (B1a), playing a pivotal role in T-cell and B-cell homeostasis and immune tolerance.
Related references: PMID:35720357 -
It mediates contact-dependent T-cell help for B-cell activation by engaging ligands such as CD5L or CD72, and maintains regulatory T and B-cell balance.
Related references: PMID:24950378, PMID:9064341, PMID:9723705 -
During thymocyte development, CD5 functions as a negative regulator of TCR signaling, associating with LCK, CD3ζ, PI3K, and CBL to modulate signal strength.
Related references: PMID:11157848 -
Co-engagement of CD3 with CD5 enhances CBL recruitment, promoting VAV1 phosphorylation and degradation, which dampens downstream TCR signaling.
-
In B-cells, CD5 modulates IL-10 production through a Ca²⁺-dependent, TRPC1-mediated pathway leading to ERK1/2 activation.
-
CD5 is a type I transmembrane protein with a predicted molecular weight of ~54 kDa, localized at the cell membrane. Post-translational modifications include disulfide bonds, glycosylation, and phosphorylation, consistent with its receptor and signaling functions.
Experimental Guidance and Technical Tips
- The immunogen comprises the extracellular domain of mouse CD5 (amino acids 25–371), providing excellent recognition of the native receptor on live cells for flow cytometry applications.
- As a directly conjugated PE antibody, it is ready-to-use in multicolor flow cytometry panels, eliminating the need for secondary staining.
- Titrate the antibody to determine the optimal staining index for your specific cell type and experimental conditions, and validate in the appropriate murine tissue or cell samples.
