Biological Background: Receptor-type tyrosine-protein phosphatase C Function and Localization
- Protein identity and alternative names: Receptor-type tyrosine-protein phosphatase C (PTPRC), also known as leukocyte common antigen (L-CA), lymphocyte antigen 5 (Ly-5), T200, and CD45. It is a transmembrane glycoprotein expressed on all nucleated hematopoietic cells.
- Core function in T-cell activation: Functions as a protein tyrosine phosphatase required for T-cell activation through the antigen receptor. Acts as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second modulates substrate specificity.
- Phosphatase activity and substrate interactions: Recruits and dephosphorylates SKAP1 and FYN upon T-cell activation, and dephosphorylates LYN, thereby modulating LYN activity.
