Biological Background: Receptor-type tyrosine-protein phosphatase C Function and Localization

  • Protein identity and alternative names: Receptor-type tyrosine-protein phosphatase C (PTPRC), also known as leukocyte common antigen (L-CA), lymphocyte antigen 5 (Ly-5), T200, and CD45. It is a transmembrane glycoprotein expressed on all nucleated hematopoietic cells.
  • Core function in T-cell activation: Functions as a protein tyrosine phosphatase required for T-cell activation through the antigen receptor. Acts as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second modulates substrate specificity.
  • Phosphatase activity and substrate interactions: Recruits and dephosphorylates SKAP1 and FYN upon T-cell activation, and dephosphorylates LYN, thereby modulating LYN activity. Also interacts with CLEC10A on dendritic cells, which recognizes Tn antigen-carrying CD45 on effector T cells to limit autoreactivity.
  • Gene family and homologs: Belongs to the protein tyrosine phosphatase family. The human ortholog is encoded by PTPRC (GeneID: 5788), and the mouse gene Ptprc (Ly-5) maps to chromosome 1.
  • Subcellular localization: Localizes to the cell membrane, membrane rafts, and the immunological synapse, consistent with its role in modulating signaling at the T-cell–APC interface.
  • Post-translational modifications and structural features: Heavily glycosylated and phosphorylated; contains alternatively spliced exons that generate multiple isoforms with molecular weights ranging from approximately 130 to 144 kDa (predicted from UniProt: 144837 Da).
  • Tissue expression: Expressed specifically in leukocytes; distinct isoform expression patterns are associated with different hematopoietic lineages and activation states.
  • Antimicrobial and other roles: While primarily a signaling molecule, CD45 plays a critical role in setting the threshold of T-cell activation and maintaining immune homeostasis.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 26–566 of mouse CD45, covering the extracellular domain. This region is critical for isoform variability; consider verifying reactivity against the desired isoform in your sample context.
  • This antibody is recommended for flow cytometry (FC) applications. For optimal results, titrate the antibody and validate in the relevant mouse cell population (e.g., splenocytes, peripheral blood lymphocytes).
  • Since CD45 is a pan-leukocyte marker, use appropriate gating strategies to distinguish leukocyte subsets. Co-staining with lineage-specific markers is advised.
  • The antibody is provided as a PE conjugate. Ensure compatibility with your cytometer’s laser and filter set (PE typically emits at ~578 nm). Protect from light and handle according to recommended ice-bag shipping conditions.
