Biological Background: CD44 antigen Function and Localization

  • CD44 antigen, also known as Ly-24, Pgp-1, HERMES, extracellular matrix receptor III (ECMR-III), GP90 lymphocyte homing/adhesion receptor, HUTCH-I, Hermes antigen, hyaluronate receptor, lymphocyte antigen 24, phagocytic glycoprotein 1 (PGP-1), and phagocytic glycoprotein I (PGP-I), is a multifunctional cell-surface glycoprotein encoded by the Cd44 gene in mouse.
  • CD44 engages extracellular matrix components like hyaluronan (HA), collagen, growth factors, cytokines, and proteases through its ectodomain, and assembles protein complexes via its cytoplasmic domain to facilitate signal transduction, influencing cell migration, adhesion, and activation.   • Upon LGALS9 ligand binding, CD44 activates downstream signaling components LCK, ERK, and MAPK to promote NK cell activation. Related references: PMID:37006235
  • CD44 participates in T-lymphocyte activation, recirculation and homing, hematopoiesis, inflammation, and response to bacterial infection.
  • Subcellularly, CD44 localizes to the cell membrane, cell projections (microvilli), and can be secreted.
