Biological Background: Leukosialin (CD43) Function and Localization
- Leukosialin (CD43, sialophorin) is a major transmembrane sialoglycoprotein on leukocytes, characterized by a signal peptide, a large extracellular mucin-like domain, a single transmembrane helix, and a cytoplasmic tail. It is heavily O-glycosylated and serves as the predominant cell surface sialoprotein.
- Regulates multiple T-cell functions including activation, proliferation, differentiation, trafficking, and migration. Positively regulates lymph-node trafficking via association with ERM proteins (EZR, RDX, MSN) (PubMed:11728336, PubMed:17638845, PubMed:21289089). - Use appropriate isotype controls and validate with known positive (e.g., mouse splenocytes or T-cell lines) and negative (e.g., CD43 knockout cells or non-hematopoietic cells) controls.
- Consider that CD43 may be shed or internalized upon activation; optimize staining time and temperature to capture surface expression accurately.
