- CD34, encoded by the Cd34 gene, is a single-pass type I membrane glycoprotein with a calculated molecular weight of approximately 41 kDa (UniProt: Q64314). Several alternatively spliced isoforms exist.
- Functions as a possible adhesion molecule implicated in early hematopoiesis, mediating stem cell attachment to the bone marrow extracellular matrix or stromal cells.
- May act as a scaffold for lineage-specific glycans, facilitating binding to lectins on stromal cells or other marrow components, and presents carbohydrate ligands to selectins (By similarity).
- Subcellular localization: membrane; contains a signal peptide and a transmembrane domain.
- Post-translational modifications include phosphorylation and N-linked glycosylation; classified as a glycoprotein and phosphoprotein.
- Tissue specificity: highly expressed in hematopoietic progenitor cell lines, brain, and testis; moderate expression in thymus, spleen, and bone marrow; not detected in adult liver. Also expressed in kidney (thin limb of Henle's loop at protein level).
