Biological Background: CD32 Function and Localization
- CD32, also known as low affinity immunoglobulin gamma Fc region receptor II (FcγRIIB), is a receptor for the Fc region of complexed IgG. It plays a key role in effector and regulatory functions, including phagocytosis of antigen-antibody complexes and modulation of antibody production by B-cells.
- Multiple isoforms are produced by alternative splicing: IIB1, IIB1', IIB2, and IIB3. Isoforms IIB1 and IIB1' can form caps but do not mediate endocytosis or phagocytosis, while isoform IIB2 mediates endocytosis of soluble immune complexes via clathrin-coated pits.
- CD32 belongs to the immunoglobulin gene family and contains Ig-like domains in its extracellular region. It acts as an inhibitory receptor when co‑ligated with BCR, TCR, or FcεRI, down‑regulating lymphocyte and mast cell activation.
- Subcellular localization includes the cell membrane, cytoplasm, cytoskeleton, and secreted forms. The different isoforms exhibit distinct trafficking behaviors, with IIB3 being released by macrophages and found in serum.
- CD32 is widely expressed on cells of hemopoietic origin. Isoform expression is lineage‑specific: IIB1 is preferentially expressed in lymphoid cells, IIB2 in myeloid cells, and IIB1' in both lineages and spleen cells but not in mesenteric lymph node cells.
