Biological Background: C-type lectin domain family 10 member A (CLEC10A) Function and Localization
- CLEC10A, also known as macrophage galactose-type lectin (MGL) or CD301, is a transmembrane C-type lectin receptor encoded by the Clec10a gene in mice.
- It specifically binds terminal galactose and N-acetylgalactosamine (GalNAc) residues, mediating interactions with glycoproteins on tumor cells and other targets.
- Plays a role in the recruitment of inflammatory monocytes to adipose tissue during diet-induced obesity, contributing to adipose tissue inflammation. Related references: PMID:19995956
- Contributes to host resistance against Trypanosoma cruzi infection by promoting optimal macrophage activation. Related references: PMID:25170304
- Localized on the cell membrane, expressed on activated macrophages and detected in various tissues including lymph node, spleen, lung, and heart.
- The antibody recognizes both CLEC10A (MGL1) and the closely related CLEC10A-like (MGL2) due to sequence homology (SWISS P49300 and Q8JZN1).
- Post-translational modifications include N-linked glycosylation and disulfide bond formation typical of C-type lectins.
Experimental Guidance and Technical Tips
- The antibody is supplied as a PE-conjugate, optimized for direct flow cytometry; no secondary staining is required.
- Because the target is a cell-surface receptor, standard surface staining protocols without permeabilization are suitable for viable cell analysis.
- Titrate the antibody to determine the optimal concentration for your specific cell type; a starting range of 0.5-5 µg per 10^6 cells is often tested.
- The immunogen covers amino acids 57-304, corresponding to the extracellular domain of CD301a, ensuring recognition of the native surface protein.
- Validate reactivity in mouse macrophage cell lines such as RAW 264.7, or in primary activated macrophages, as the protein is expressed on activated myeloid cells.
- For studying MGL1 vs MGL2, confirm isoform specificity with additional assays since this pan-CD301 antibody recognizes both.
- Include appropriate isotype controls and Fc blocking steps to minimize non-specific binding in flow cytometry.
