Biological Background: CD19 Function and Localization
- CD19 is a B-lymphocyte antigen that functions as a co-receptor for the B-cell antigen receptor (BCR) complex, lowering the threshold for B-cell activation.
- Upon engagement, CD19 activates phosphatidylinositol 3-kinase and mobilizes intracellular Ca²⁺ stores, driving downstream signaling cascades. Related references: PMID:12387743, PMID:20101619, PMID:9382888
- CD19 is not essential for early B-cell differentiation in the bone marrow but is critical for the normal development of B-1 cells. Related references: PMID:7542548, PMID:7543183, PMID:9317126, PMID:12387743
- It is required for normal B-cell proliferation and differentiation in response to antigen challenge, and for the production of high-affinity antibodies and normal serum immunoglobulin levels. Related references: PMID:12387743, PMID:7542548, PMID:7543183, PMID:9317126
- CD19 localizes to the cell membrane and is enriched in membrane rafts.
- It is detected on B cells in spleen, bone marrow, thymus, lymph nodes, and peripheral blood lymphocytes (at protein level). Related references: PMID:12387743, PMID:20101619, PMID:7542548, PMID:7543183
- CD19 is a transmembrane glycoprotein containing immunoglobulin-like domains, disulfide bonds, and phosphorylation sites, belonging to the immunoglobulin superfamily.
Experimental Guidance and Technical Tips
- The PE-conjugated format allows direct detection in flow cytometry, eliminating the need for a secondary antibody staining step.
- Consider titrating the antibody to determine the optimal concentration for your specific cell staining protocol and experimental conditions.
- Validate the antibody in your target tissue or cell line, as CD19 expression levels may vary; include appropriate isotype controls and Fc blocking to minimize non-specific binding.
- For multicolor flow cytometry panels, check fluorochrome compatibility to avoid spectral overlap issues.
