Biological Background: CD18/Integrin beta-2 Function and Localization
- Integrin beta-2 (CD18) pairs with alpha subunits (CD11a, CD11b, CD11c, CD11d) to form leukocyte-specific integrins: LFA-1 (αLβ2), Mac-1 (αMβ2), p150,95 (αXβ2), and αDβ2. These receptors mediate cell adhesion and signaling.
- LFA-1 binds ICAM-1, -2, -3, -4 and ISG15; Mac-1 and p150,95 bind iC3b, fibrinogen, and factor X; αDβ2 binds ICAM-3 and VCAM-1; contributing to immune cell adhesion, migration, and phagocytosis.
- CD18 is crucial for leukocyte transmigration; for example, neutrophil recruitment during lung injury requires integrin signaling via PTK2B/PYK2 (PubMed:18587400). Related references: PMID:18587400
- Plays a role in natural killer cell cytotoxicity and phagocytosis of apoptotic neutrophils by macrophages (via LFA-1/ICAM-3).
- Mac-1 (ITGAM/ITGB2) is critical for mast cell development and in immune complex-mediated glomerulonephritis; ITGAM-null mice show neutrophil accumulation, impaired degranulation, and phagocytosis.
- Also functions in erythrocyte adhesion and phagocytosis (ICAM4) and neuronal adhesion (ICAM5).
- Subcellular localization: Cell membrane, enriched in membrane rafts.
- Post-translational modifications: Glycoprotein, disulfide bonds, phosphorylation, and contains EGF-like domains; requires calcium and magnesium for function.
Experimental Guidance and Technical Tips
- This PE-conjugated rabbit monoclonal antibody is designed for flow cytometry (FC). For optimal results, titrate the antibody to determine the ideal working concentration for your specific cell type and assay conditions.
- Use appropriate isotype controls and viable cell gating to ensure specific staining. Protect the antibody from prolonged light exposure and avoid fixatives that may quench PE fluorescence.
- The immunogen spans the extracellular domain (aa 1-771) of mouse CD18, suggesting recognition of native conformation. Validate cross-reactivity if using non-mouse species, as this product is reported to react with mouse only.
- For multi-color flow panels, verify fluorophore compatibility and perform compensation controls.
