Biological Background: CD150/SLAM Function and Localization
- Encodes signaling lymphocytic activation molecule (SLAM), also known as SLAM family member 1 (Slamf1), a self-ligand receptor of the SLAM family that modulates immune cell activation and differentiation through homo- or heterotypic cell-cell interactions.
- In T-cells, mediates IL-2-independent proliferation and induces IFN-gamma production. Related references: PMID:12351401, PMID:9126961
- Downstream signaling involves INPP5D, DOK1, and DOK2 leading to inhibited IFN-gamma production, as well as PRKCQ, BCL10, and NFKB1 leading to increased T-cell activation and Th2 cytokine production. Related references: PMID:11477403, PMID:15539155, PMID:16847311
- Promotes TCR-induced IL-4 secretion by CD4+ T-cells and is required for IL-4 production by germinal center T follicular helper cells. Related references: PMID:15123745, PMID:20525889
- In B-cells, activates ERK and Akt signaling pathways independently of SH2D1A, with downstream signals implicating SYK and INPP5D. Related references: PMID:15315965
- Functions in innate immunity by recognizing Gram-negative bacteria in macrophages, regulating phagosome maturation, and promoting NOX2 activity. Related references: PMID:20818396, PMID:22493499
- Localized to the cell membrane as a single-pass type I transmembrane protein, with an extracellular region containing immunoglobulin-like domains.
- Post-translationally modified by glycosylation and phosphorylation, playing roles in cell adhesion, signaling, and both adaptive and innate immunity.
Experimental Guidance and Technical Tips
- The immunogen spans the extracellular domain (aa 25–242), making this PE-conjugated antibody ideal for detecting cell surface CD150/SLAM via flow cytometry.
- For flow cytometry, titrate the antibody to determine the optimal staining concentration for your specific cell type and experiment.
- Ensure appropriate isotype controls are included to discriminate specific staining.
- The antibody may also be suitable for other applications such as immunohistochemistry or functional studies; however, these have not been validated and would require careful optimization.
