Biological Background: CD144/VE-Cadherin Function and Localization

  • Cadherin-5 (CD144, VE-cadherin), encoded by the Cdh5 gene in mouse, is a classical cadherin cell adhesion molecule.
  • This cadherin may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions. Related references: PMID:20332120 PMID:9220534
  • It associates with alpha-catenin forming a link to the cytoskeleton, and acts as a cell junctional complex anchor for AMOTL2 and MAGI1 to couple actin fibers to cell junctions. Related references: PMID:24806444
  • Acts in concert with KRIT1 and PALS1 to establish and maintain correct endothelial cell polarity and vascular lumen, mediated by recruitment and activation of the Par polarity complex and RAP1B. Related references: PMID:27466317
  • Associates with CTNND1/p120-catenin to control CADH5 endocytosis. Related references: PMID:33972531
  • Subcellularly, it localizes to adherens junctions, cell membrane, and cytoplasm.
  • Expressed in postnatal endothelial cells of the retinal vascular plexus (at protein level).
  • Annotated with keywords including calcium-binding, glycoprotein, phosphoprotein, and transmembrane.

Experimental Guidance and Technical Tips

  • The immunogen comprises amino acids 46-592 of mouse VE-cadherin, covering a large portion of the extracellular region including cadherin repeats, which may allow detection of the native cell surface protein.
  • This antibody is conjugated to PE for direct detection in flow cytometry; a corresponding isotype control and Fc receptor blocking are recommended for optimal results.
  • Target validation is advised in known VE-cadherin-expressing endothelial cells (e.g., HUVECs, bEnd.3, or primary mouse endothelial cells) to confirm reactivity.
  • For fixation and permeabilization protocols, assess compatibility with the PE fluorophore, as some conditions may affect fluorescence.
  • As this antibody is validated for mouse, cross-reactivity with other species should be verified if intended for multi-species studies.
