Biological Background: Interleukin-6 Receptor Subunit Beta (IL6ST/gp130) Function and Localization

  • IL6ST (gp130) serves as the shared signal-transducing receptor subunit for the IL-6 family of cytokines, including IL-6, LIF, OSM, CNTF, IL-11, CTF1, and CLCF1. Cytokine binding triggers homodimerization (with IL-6 or IL-11) or heterodimerization (with LIFR or OSMR) to initiate downstream signaling. Related references: PMID:1602143, PMID:10661409

  • Tyrosine kinases of the JAK family are constitutively associated with the intracellular domains of gp130. Ligand-induced dimerization activates JAKs, which phosphorylate gp130, creating docking sites for STAT transcription factors (mainly STAT3) to regulate gene expression. Related references: PMID:10661409, PMID:1602143

  • The IL-6 signaling pathway induces expression of SOCS1 and SOCS3, which directly inhibit JAK activity and serve as a negative feedback loop to terminate signaling. Related references: PMID:9202125

  • Beyond STAT3, gp130 can activate the YAP and NOTCH pathways to promote inflammation-induced epithelial regeneration, independent of STAT3. Related references: PMID:25731159

  • gp130 is essential for a broad range of physiological processes, including immune regulation, hematopoiesis, pain perception, bone metabolism, and embryonic development, as demonstrated in gene-targeted mice. Related references: PMID:10661409, PMID:25057188, PMID:26255596

  • It is required for the survival of motor and sensory neurons, differentiation of astrocytes, and osteoblast differentiation via maintenance of PTH1R expression, contributing to normal trabecular bone mass and cortical bone composition. Related references: PMID:10377352, PMID:25228504, PMID:24339143, PMID:9348227

  • gp130 is a cell membrane glycoprotein with an extracellular immunoglobulin domain. It is widely expressed across tissues, including brain (paraventricular nucleus), heart, thymus, spleen, kidney, lung, and liver, and in most cell lines tested. Related references: PMID:28402851

  • The mature protein has a predicted molecular weight of approximately 102 kDa, consistent with its glycosylation state.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (amino acids 23–617) of mouse gp130, which encompasses the cytokine-binding and immunoglobulin-like regions; this design may favor recognition of the native conformation on live cells.
  • This PE-conjugated antibody is intended for flow cytometry (FC). For optimal staining, titrate the antibody and include appropriate isotype controls. Fixation and permeabilization steps could alter epitope accessibility; always validate in the relevant sample system.
  • Cross-reactivity has been demonstrated with mouse. When applying to other species, verify reactivity experimentally.
