Biological Background: Integrin alpha-M Function and Localization
- Integrin alpha-M (ITGAM) is a member of the integrin alpha chain family, characterized by multiple FG-GAP repeats and an I domain, and exists in several isoforms due to alternative splicing.
- Forms a heterodimer with ITGB2 (CD18) to create the complement receptor 3 (CR3) that binds iC3b-opsonized particles and recognizes the R-G-D peptide in C3b, mediating phagocytosis.
- Functions as an adhesion receptor for fibrinogen, factor X, and ICAM1, facilitating leukocyte adhesion, migration, and neutrophil–endothelial interactions.
- In microglia, the ITGAM/ITGB2 complex associates with TYROBP/DAP12 to regulate superoxide production, which influences neuronal apoptosis during brain development. Related references: PMID:18685038
- Predominantly expressed on monocytes, granulocytes, peritoneal mast cells, and microglia. Related references: PMID:3887182, PMID:8986723, PMID:9862668, PMID:18685038
- Localized to the cell membrane and membrane rafts; contains structurally essential disulfide bonds, glycosylation sites, and calcium-binding domains.
- Possesses a signal peptide, a large extracellular domain, a single transmembrane helix, and a short cytoplasmic tail.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 143–337 of the extracellular domain, a region likely surface-exposed; the antibody is suitable for flow cytometry of live, unfixed mouse cells.
- For intracellular staining or protocols involving fixation/permeabilization, confirm that the target epitope remains intact under the chosen conditions.
- Titrate the antibody in your specific experimental system, and include appropriate isotype controls to ensure staining specificity.
- CD11b is expressed on mouse monocytes, granulocytes, and microglia; use relevant cell lines (e.g., J774, RAW 264.7) or primary cells as positive controls.
