Biological Background: CCR6 Function and Localization
- CCR6 (C-C chemokine receptor type 6), also designated KY411 or Cmkbr6, is a G protein-coupled receptor with a molecular weight of approximately 42 kDa.
- CCR6 is the receptor for chemokine CCL20 and also binds beta-defensins (DEFB1, DEFB4, DEFB4A/B), mediating chemotactic and signaling effects (PubMed:20068036, PubMed:25122636).
Related references: PMID:20068036, PMID:25122636 - The CCL20-CCR6 axis drives chemotaxis of dendritic cells, effector/memory T-cells, and B-cells, and is essential for immune responses at mucosal surfaces and under inflammatory conditions (PubMed:21376174).
Related references: PMID:21376174 - CCR6 is critical for recruiting pro-inflammatory Th17 cells and regulatory T-cells (Tregs) to inflammation sites, and for normal Th17 migration in intestinal tissues (PubMed:19050256, PubMed:19129757).
Related references: PMID:19050256, PMID:19129757 - It contributes to early thymocyte precursor migration and optimal DN2/DN3 thymocyte differentiation (PubMed:24638065).
Related references: PMID:24638065 - CCR6 is essential for B-cell localization in Peyers-patches subepithelial dome, IgA class switching, and memory B-cell distribution and recall responses in the spleen (PubMed:27174992, PubMed:25505290).
Related references: PMID:27174992, PMID:25505290 - In sperm, CCR6 positively regulates motility and chemotaxis via CCL20 signaling and is predominantly localized to the principal piece and neck of the tail (PubMed:23765988).
Related references: PMID:23765988 - CCR6 localizes to the cell membrane and is expressed in testis, lung, colon, dendritic cells, and lymphocyte subsets including Th17, Treg, and memory B cells.
Experimental Guidance and Technical Tips
- The immunogen (recombinant fusion protein spanning amino acids 2-38 of the mouse CCR6 N-terminus) targets the extracellular domain, which may allow recognition of native cell surface receptor in flow cytometry.
- This PE-conjugated monoclonal antibody is designed for direct flow cytometry (FC); optimize titration in your specific sample matrix and assess potential Fc receptor blocking.
- For mouse studies, consider testing on tissues or cell types with known CCR6 expression, such as Peyer's patches, lamina propria lymphocytes, or in vitro-differentiated Th17 cells.
