Biological Background: FOXP3 Function and Localization

  • FOXP3 (Forkhead box protein P3), also known as scurfin, is a transcription factor critical for the development and suppressive function of regulatory T-cells (Tregs). Related references: PMID:17377532, PMID:21458306, PMID:23947341, PMID:23169781

  • Acts as both a transcriptional repressor and activator, modulating key immune genes such as CTLA4, TNFRSF18, IL2, and IFNG to maintain immune homeostasis. Related references: PMID:17377532, PMID:21458306, PMID:23947341

  • Mediates repression of IL2 through association with histone acetylase KAT5 and histone deacetylase HDAC7. Related references: PMID:17360565

  • Inhibits cytokine production by repressing the activity of transcription factors RELA and NFATC2. Related参考文献: PMID:15790681

  • Interacts with RUNX1 to regulate expression of CTLA4, IL2, and IFNG. Related references: PMID:17377532

  • Antagonizes RORC and RORA, thereby inhibiting Th17 differentiation and promoting Treg development. Related references: PMID:18368049, PMID:18354202

  • Localizes predominantly in the nucleus, with some cytoplasmic presence.

  • Mutations in FOXP3 cause IPEX (Immune dysregulation, Polyendocrinopathy, Enteropathy, X-linked) syndrome, characterized by severe autoimmunity.

Experimental Guidance and Technical Tips

  • For intracellular flow cytometry, use appropriate fixation and permeabilization buffers optimized for nuclear antigens.
  • Titrate the antibody to determine the optimal staining concentration for your specific cell type and experimental conditions.
  • Validate species cross-reactivity with positive control cells or tissues from human, mouse, and cynomolgus.
  • Include an isotype control and fluorescence-minus-one (FMO) controls to ensure specific staining.
  • For Treg analysis, co-stain with CD4 and CD25 to identify the classical Treg population.
