Biological Background: Integrin alpha-5 Function and Localization
- ITGA5 pairs with ITGB1 to form the classic fibronectin receptor (VLA-5), which recognizes the R-G-D tripeptide sequence in matrix ligands.
- ITGA5:ITGB1 mediates R-G-D-dependent cell adhesion to fibronectin (FN1). Related references: PMID:33962943
- ITGA5:ITGB1 also binds to fibrillin-1 (FBN1), facilitating cell attachment to elastic fibers. Related references: PMID:12807887, PMID:17158881
- The integrin interacts with PLA2G2A at a distinct site, inducing conformational changes that enhance ligand-binding affinity. Related references: PMID:18635536, PMID:25398877
- ITGA5:ITGB1 acts as a receptor for IL1B, and ITGA5:ITGB3 serves as a receptor for soluble CD40LG. Related references: PMID:29030430, PMID:31331973
- Beyond adhesion, ITGA5 participates in viral entry: ITGA5:ITGB1 is a receptor for Human metapneumovirus and Human parvovirus B19; in HIV-1, the viral Tat protein binds ITGA5:ITGB1, enhancing angiogenesis in Kaposi's sarcoma.
- Subcellularly, ITGA5 localizes to the cell membrane and focal adhesions; it requires calcium/metal-ion binding and undergoes phosphorylation and glycosylation.
- The protein is expressed in placenta (at the protein level) according to UniProt tissue specificity.
Experimental Guidance and Technical Tips
- The immunogen (aa 42–995) covers nearly the entire extracellular domain, suggesting the antibody likely recognizes native ITGA5 on live cells. This supports its use in flow cytometry (FC) applications as indicated.
- For FC, a recommended starting concentration is typically 0.5–5 µg/mL, but optimal titrations should be determined experimentally using cells with known ITGA5 expression (e.g., HUVEC, NIH/3T3) and appropriate isotype controls.
- The PE conjugate allows direct detection without a secondary antibody, reducing background and simplifying multicolor panel design. Validate instrument compensation settings with positive and negative cell populations.
- Cross-reactivity with mouse ITGA5 enables translational studies; confirm reactivity in your specific mouse strain or cell line.
