Biological Background: CD69 Function and Localization
- Early activation antigen CD69 (also known as AIM, EA1, MLR-3, CLEC2C, BL-AC/P26, GP32/28, Leu-23) is a type II transmembrane C-type lectin receptor.
- Rapidly upregulated on the surface of activated T-cells, B-cells, NK cells, neutrophils, eosinophils, and platelets; serves as a key marker for immune cell activation and homeostasis.
- Negatively regulates Th17 cell differentiation through carbohydrate-dependent interaction with galectin-1/LGALS1 present on immature dendritic cells (PubMed:24752896). Related references: PMID:24752896
- Promotes conversion of naive CD4 T-cells into regulatory T-cells via interaction with the S100A8/S100A9 complex on peripheral blood mononuclear cells (PubMed:26296369). Related references: PMID:26296369
- Acts as an oxidized low-density lipoprotein (oxLDL) receptor in CD4 T-lymphocytes, negatively regulating inflammatory responses by inducing PDCD1 expression through NFAT activation (PubMed:35930205). Related references: PMID:35930205
- Participates in antibacterial protection by specifically recognizing P. aeruginosa, which triggers ERK1 activation and granulocyte-macrophage colony-stimulating factor (GM-CSF) secretion (PubMed:34841721). Related references: PMID:34841721
- Negatively regulates chemotactic responses of effector lymphocytes and dendritic cells by acting as an S1PR1 receptor agonist, facilitating receptor internalization and degradation (PubMed:37039481). Related references: PMID:37039481
- Subcellular localization: cell membrane; post-translational modifications include disulfide bonds, glycosylation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen (aa 78–199) corresponds to the predicted extracellular domain, suggesting the antibody recognizes cell surface CD69, making it suitable for direct flow cytometry on live cells.
- Since CD69 expression is activation-dependent, consider verifying cell stimulation conditions and time course in your experimental system.
- Titrate the PE-conjugated antibody to determine the optimal staining concentration for your cell type and application; include appropriate isotype controls and viability dyes.
- Standard FC protocols with single-cell suspensions are recommended; avoid procedures that may internalize or shed surface antigens.
