Biological Background: CD45 Function and Localization
- CD45, also known as leukocyte common antigen (L-CA) and T200, is a receptor-type tyrosine-protein phosphatase C encoded by the PTPRC gene.
- It is essential for T-cell activation via the antigen receptor, acting as a positive regulator of coactivation upon DPP4 binding.
Related references: PMID:35767951 - The protein contains two phosphatase domains; the first has enzymatic activity, and the second modulates substrate specificity. It dephosphorylates key signaling molecules such as SKAP1, FYN, and LYN.
- CD45 is a single-pass type I membrane protein located in membrane rafts and at the immunological synapse.
- Multiple isoforms (1–8) are generated by alternative splicing, with tissue-specific expression; isoforms 1,2,3,5,7 are detected in thymocytes.
- It plays a role in modulating immune signaling, including interaction with CLEC10A to limit autoreactivity, and can act as a receptor for human cytomegalovirus UL11, impairing T-cell proliferation.
- Post-translational modifications include glycosylation and phosphorylation; the protein belongs to the protein phosphatase family and is linked to severe combined immunodeficiency (SCID) disease variants.
Experimental Guidance and Technical Tips
- This PE‑conjugated monoclonal antibody is optimized for direct flow cytometry detection of surface CD45 on live cells. Consider titrating the antibody to determine the optimal concentration for your experimental system.
- Include appropriate isotype controls and fluorescence-minus-one (FMO) controls to ensure specificity.
- The immunogen covers the extracellular region (aa 26–577), so the antibody recognizes surface‑expressed CD45. Confirm surface staining using a viability dye to exclude dead cells.
- Cross‑reactivity with cynomolgus CD45 has been reported; validate performance in the relevant non‑human primate samples if needed.
- CD45 isoforms have varying molecular weights (131–147 kDa); migrate accordingly in WB as a positive control, though this product is validated only for FC.
