Biological Background: CD4 Function and Localization
- Encoded by the CD4 gene, T-cell surface glycoprotein CD4 (also known as T4, Leu-3) is an integral membrane glycoprotein with a molecular weight of approximately 51 kDa, containing immunoglobulin-like domains.
- Functions as a coreceptor for MHC class II molecules on antigen-presenting cells, interacting simultaneously with the T-cell receptor (TCR) and MHC class II to enhance T-cell activation.
- Recruits the Src kinase LCK to the TCR-CD3 complex, initiating intracellular signaling cascades that promote lymphokine production, motility, adhesion, and T-helper cell activation.
- In macrophages and NK cells, CD4 participates in differentiation, activation, cytokine expression, and cell migration through TCR/LCK-independent pathways; it also triggers monocyte differentiation into mature macrophages.
- Plays a critical role in thymic development of T-helper cells and is highly expressed on T-helper cells, with additional presence on macrophages, dendritic cells, and NK cells.
- Serves as the primary receptor for human immunodeficiency virus-1 (HIV-1), with viral entry mediated by gp120 binding; additionally acts as a receptor for Human Herpesvirus 7 (HHV-7). - Subcellularly localized to the cell membrane, CD4 is subject to post-translational modifications including disulfide bond formation, glycosylation, palmitoylation, and phosphorylation, which regulate its function and interactions.
