Biological Background: CD14 Function and Localization

  • CD14, also known as monocyte differentiation antigen, My23 antigen, or myeloid cell-specific leucine-rich glycoprotein, is a key player in innate immunity.
  • It acts as a coreceptor for bacterial lipopolysaccharide (LPS), binding monomeric LPS in concert with LBP and delivering it to the LY96/TLR4 complex, thereby mediating the innate immune response to bacterial LPS. Related references: PMID:1698311 PMID:23264655 PMID:20133493 PMID:22265692
  • Downstream signaling occurs via MyD88, TIRAP, and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and the inflammatory response. Related references: PMID:8612135
  • CD14 also functions as a coreceptor for TLR2:TLR6 and TLR2:TLR1 heterodimers in response to diacylated and triacylated lipopeptides; these clusters trigger cell surface signaling and subsequent lipid-raft-dependent trafficking to the Golgi. Related references: PMID:16880211
