Biological Background: TIGIT Function and Localization
- TIGIT (T-cell immunoreceptor with Ig and ITIM domains) is an inhibitory receptor of the immunoglobulin superfamily, also known as VSIG9 and VSTM3, that modulates immune responses principally by suppressing T-cell activation through promoting mature immunoregulatory dendritic cells. - It binds with high affinity to the poliovirus receptor PVR/CD155 and nectin-2/CD112 on antigen-presenting cells, delivering inhibitory signals to T-cells and NK cells; ligand engagement triggers phosphorylation of its cytoplasmic tail by Src kinases FYN or LCK, recruiting GRB2 and the phosphatase SHIP1 to dampen PI3K and MAPK pathways.
