Biological Background: TIGIT Function and Localization

  • TIGIT (T-cell immunoreceptor with Ig and ITIM domains) is an inhibitory receptor of the immunoglobulin superfamily, also known as VSIG9 and VSTM3, that modulates immune responses principally by suppressing T-cell activation through promoting mature immunoregulatory dendritic cells. Related references: PMID:19011627
  • It binds with high affinity to the poliovirus receptor PVR/CD155 and nectin-2/CD112 on antigen-presenting cells, delivering inhibitory signals to T-cells and NK cells; ligand engagement triggers phosphorylation of its cytoplasmic tail by Src kinases FYN or LCK, recruiting GRB2 and the phosphatase SHIP1 to dampen PI3K and MAPK pathways. Related references: PMID:23154388
  • Additionally, TIGIT associates with β-arrestin-2/ARRB2 to recruit SHIP1, which suppresses autoubiquitination of TRAF6 and consequently inhibits the NF-κB signaling cascade. Related references: PMID:24817116
  • It also acts as a functional receptor for nectin-4 to inhibit NK cell cytotoxicity, expanding its immune checkpoint repertoire. Related references: PMID:32503945
  • At the protein level, TIGIT is expressed at low levels on peripheral memory and regulatory CD4+ T-cells and NK cells and is markedly up-regulated following activation of these cells.
  • Subcellularly, it localizes to the cell membrane as a single-pass type I transmembrane protein.
  • Post-translational modifications include glycosylation, disulfide bond formation, and phosphorylation, which are essential for its signaling functions.
  • Alternative splicing generates isoforms of the receptor, and the protein is a member of the reference proteome with a calculated molecular mass of approximately 26.3 kDa.

Experimental Guidance and Technical Tips

  • This PE-conjugated rabbit monoclonal antibody is designed for direct flow cytometry (FC) detection of human TIGIT on live cells; no secondary staining is required.
  • Given the recombinant immunogen spanning the extracellular domain (amino acids 22–141), the antibody likely recognizes an epitope within the ligand-binding region; consider performing blocking experiments with PVR/CD155 or nectin-2 to confirm specificity.
  • Since TIGIT expression is low on resting cells and upregulated upon activation, include an appropriate activation step (e.g., anti-CD3/CD28 stimulation for T cells or IL-2 for NK cells) to maximize signal.
  • Titrate the antibody to determine the optimal working concentration for your specific cell type and assay conditions, and always include appropriate isotype controls.
