Biological Background: STAT3 Function and Localization

  • STAT3 (Signal transducer and activator of transcription 3), also known as acute-phase response factor (APRF), is a cytoplasmic transcription factor that mediates cellular responses to interleukins, KITLG/SCF, LEP, and other growth factors (PubMed:10688651, PubMed:11294841, PubMed:12359225). Related references: PMID:10688651 PMID:11294841 PMID:12359225

  • Upon activation, STAT3 recruits transcriptional coactivators such as NCOA1 and MED1 to target gene promoters, modulating gene expression (PubMed:15653507, PubMed:16285960, PubMed:17344214). Related references: PMID:15653507 PMID:16285960 PMID:17344214

  • STAT3 regulates naive CD4+ T-cell differentiation into Th17 or Treg subsets, with acetylation enhancing its transcriptional activity and deacetylation/oxidation by LOXL3 suppressing it (PubMed:28065600, PubMed:28262505). Related references: PMID:28065600 PMID:28262505

  • STAT3 promotes cell cycle progression from G1 to S phase by inducing CCND1 expression (PubMed:17344214) and may transactivate the survival gene BIRC5 under leptin (LEP) signaling (PubMed:18242580). Related references: PMID:17344214 PMID:18242580

  • Cytoplasmic STAT3 represses macroautophagy through inhibition of EIF2AK2/PKR activity (PubMed:23084476). Related references: PMID:23084476

  • STAT3 is widely expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas, as well as in naive CD4+ T cells and T-helper Th17, Th1, and Th2 cells (PubMed:31899195). It shuttles between the cytoplasm and nucleus. Related references: PMID:31899195

  • STAT3 is involved in oncostatin-M-mediated signaling through both type I (LIFR-IL6ST) and type II (OSMR-IL6ST) receptor complexes (PubMed:9188471). Related references: PMID:9188471

  • May mediate cellular responses to activated FGFR1, FGFR2, FGFR3, and FGFR4 (PubMed:12873986) and is activated by IL31 via IL31RA (PubMed:15194700). Related references: PMID:12873986 PMID:15194700

Experimental Guidance and Technical Tips

  • For intracellular flow cytometry, ensure proper fixation and permeabilization protocols are used to allow antibody access to cytoplasmic and nuclear STAT3.
  • The immunogen targets the C-terminal region (aa 611–710); this antibody detects total STAT3. For activation state-specific detection (e.g., phospho-STAT3), use phospho-specific antibodies in parallel or employ appropriate cytokine stimulation (e.g., IL-6) prior to staining.
  • PE conjugation provides bright fluorescence; remember to include appropriate compensation controls and single-stain references in multicolor experiments.
  • Cross-reactivity is validated for human; if using with other species, validate specificity empirically with appropriate controls (e.g., knockout/knockdown samples).
  • Optimal working concentration should be determined by titration in the experimental system; a suggested starting concentration for FC is 0.5–5 µg/mL.
