- For intracellular flow cytometry, ensure proper fixation and permeabilization protocols are used to allow antibody access to cytoplasmic and nuclear STAT3.
- The immunogen targets the C-terminal region (aa 611–710); this antibody detects total STAT3. For activation state-specific detection (e.g., phospho-STAT3), use phospho-specific antibodies in parallel or employ appropriate cytokine stimulation (e.g., IL-6) prior to staining.
- PE conjugation provides bright fluorescence; remember to include appropriate compensation controls and single-stain references in multicolor experiments.
- Cross-reactivity is validated for human; if using with other species, validate specificity empirically with appropriate controls (e.g., knockout/knockdown samples).
- Optimal working concentration should be determined by titration in the experimental system; a suggested starting concentration for FC is 0.5–5 µg/mL.
