Biological Background: PD-L1/CD274 Function and Localization
- PD-L1 (CD274), also known as B7-H1, is a type I transmembrane protein belonging to the immunoglobulin superfamily.
- It serves as a ligand for the inhibitory receptor PD-1 (PDCD1) and plays a pivotal role in maintaining peripheral immune tolerance by limiting T-cell effector responses. Related references: PMID:11015443, PMID:28813410, PMID:28813417
- PD-L1 may also engage an unknown activating receptor to costimulate interleukin-10 (IL-10) production by T-cell subsets. Related references: PMID:10581077
- In the tumor microenvironment, PD-L1 is exploited by cancers to suppress antitumor immunity, leading to cytotoxic T-lymphocyte exhaustion. Blocking this interaction reverses T-cell dysfunction and is a key strategy in cancer immunotherapy. Related references: PMID:28813410, PMID:28813417
- Beyond its receptor function, PD-L1 can translocate to the nucleus under hypoxic conditions and act as a transcription coactivator to promote GSDMC expression, inducing pyroptosis. Related references: PMID:32929201
- Subcellularly, PD-L1 is predominantly localized on the cell membrane, but can also be found in early and recycling endosomes, the nucleus, and as a secreted form.
- It is highly expressed in the heart, skeletal muscle, placenta, and lung, and is induced on activated T cells, B cells, dendritic cells, keratinocytes, and monocytes.
- Keywords associated with PD-L1 include immunity, transmembrane, glycoprotein, disulfide bond, and alternative splicing.
Experimental Guidance and Technical Tips
- Given that the immunogen comprises the extracellular domain (amino acids 1-290), the antibody is expected to recognize the native, surface-exposed PD-L1 protein, making it suitable for flow cytometry without permeabilization.
- For FC experiments, titrate the PE-conjugated antibody in your cellular system to determine the optimal concentration for specific staining. Include proper isotype controls and live/dead discrimination.
- PD-L1 expression can vary with cell type, activation status, and microenvironmental cues (e.g., hypoxia, inflammatory cytokines). Validate detection in your specific sample context.
- Consider using this antibody for surface staining of human PBMCs, tumor cell lines, or dissociated tissue samples. The reported molecular weight of 33 kDa can serve as a reference for complementary immunoblotting, though PE conjugates are primarily optimized for flow-based readouts.
