Biological Background: NT5E/CD73 Function and Localization
- NT5E/CD73, also known as ecto-5'-nucleotidase, is a glycosylphosphatidylinositol (GPI)-anchored cell membrane protein that catalyzes the hydrolysis of extracellular nucleotide monophosphates into their corresponding nucleosides and inorganic phosphate.
- The enzyme exhibits a preference for AMP as a substrate, but can also hydrolyze IMP, UMP, GMP, CMP, and deoxyribonucleotide monophosphates such as dAMP, dCMP, and dTMP. Related references: PMID:21933152, PMID:22997138, PMID:23142347
- NT5E shows selectivity for ribonucleotide monophosphates over deoxyribose forms. Related references: PMID:34403084
- The protein is localized on the cell membrane where it regulates purinergic signaling by converting extracellular AMP to adenosine, a potent immunosuppressive metabolite.
- NT5E is a member of the 5'-nucleotidase family and requires zinc ions for its hydrolytic activity, as indicated by metal-binding domains.
- Alternative splicing of the NT5E gene produces multiple transcript variants, though the functional significance of all isoforms is not fully characterized.
- Post-translational modifications include N-linked glycosylation, disulfide bond formation, and the addition of a GPI anchor that tethers the enzyme to the cell surface.
Experimental Guidance and Technical Tips
- This PE-conjugated monoclonal antibody is designed for direct flow cytometric detection of CD73 on the surface of live human cells. Ensure cells are viable to preserve membrane integrity and optimal epitope presentation.
- The immunogen corresponds to the extracellular domain (a.a. 27-547), so the antibody is expected to recognize an epitope accessible on the cell surface without the need for permeabilization.
- For best results, titrate the antibody to determine the optimal concentration for your specific cell type and staining conditions. Typically, 0.1–1 µg per 1×10^6 cells is a reasonable starting range.
- Include appropriate isotype controls and compensation controls when performing multicolor flow cytometry to account for non-specific binding and spectral overlap.
- CD73 expression can be modulated by cellular activation; consider validating the antibody in both resting and stimulated samples, if relevant to your study.
