Biological Background: MERTK Function and Localization
- MERTK is a receptor tyrosine kinase that transduces extracellular signals by binding ligands such as LGALS3, TUB, TULP1, and GAS6.
- Upon ligand binding, MERTK autophosphorylates and interacts with GRB2 or PLCG2, leading to phosphorylation of downstream effectors including MAPK1, MAPK2, FAK/PTK2, or RAC1, thereby regulating processes like cytoskeleton reorganization and engulfment. Related references: PMID:32640697
- It plays a critical role in macrophage-mediated clearance of apoptotic cells (efferocytosis) and platelet aggregation.
- In the retinal pigment epithelium (RPE), MERTK functions as a regulator of phagocytosis of rod outer segment fragments.
- MERTK inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces production of SOCS1 and SOCS3.
- The protein is localized at the cell membrane and contains characteristic domains such as immunoglobulin domains, a kinase domain, and is subject to glycosylation and phosphorylation.
