Biological Background: LAMP1/CD107a Function and Localization

  • LAMP1 (CD107a), also known as LAMPA or LGP120, is a heavily glycosylated lysosomal membrane glycoprotein with a core molecular weight of approximately 45 kDa; due to extensive N- and O-linked glycosylation, it migrates at ~120 kDa on SDS-PAGE.
  • Functions in lysosome biogenesis, pH regulation, autophagy, and cholesterol homeostasis. Related references: PMID:37390818
  • Directly inhibits the proton channel TMEM175 to facilitate lysosomal acidification, essential for optimal hydrolase activity. Related references: PMID:37390818
  • Plays a critical role in NK cell cytotoxicity by mediating granule movement and perforin trafficking, and protects NK cells from self-damage. Related references: PMID:2022921, PMID:23632890, PMID:23847195
  • Presents carbohydrate ligands to selectins, potentially involved in cell adhesion. Related references: PMID:7685349
  • Serves as a receptor for Lassa virus glycoprotein and promotes viral-host membrane fusion. Related references: PMID:24970085, PMID:25972533, PMID:27605678, PMID:29295909
  • Supports FURIN-mediated cleavage of mumps virus fusion protein F.
  • Subcellular localization spans lysosomal, endosomal, and plasma membranes, as well as cytolytic granules.

Experimental Guidance and Technical Tips

  • For intracellular flow cytometry, ensure adequate permeabilization; LAMP1 is predominantly intracellular, so surface staining may be limited without permeabilization.
  • Validate staining in relevant cell types such as NK cells, activated T cells, or lysosome-rich cell lines to confirm expected localization.
  • Consider co-staining with lysosomal markers (e.g., LAMP2) or organelle-specific dyes to confirm lysosomal localization and assess autophagic flux.
  • The immunogen covers the luminal domain (aa 29–382), which may detect both nascent and mature forms; note that extensive glycosylation can affect antibody binding in some sample types.
