Biological Background: Integrin alpha-3 Function and Localization
- Integrin alpha-3 (UniProt: P26006, also known as CD49c, VLA-3 alpha chain, GAPB3) is a transmembrane glycoprotein that heterodimerizes with integrin beta-1 to form the alpha-3/beta-1 receptor. The alpha subunit contains a large extracellular domain with seven FG-GAP repeats, a transmembrane helix, and a short cytoplasmic tail.
- ITGA3/ITGB1 serves as a versatile adhesion receptor for multiple extracellular matrix ligands, including fibronectin, laminin, collagen, epiligrin, thrombospondin, and CSPG4, mediating cell–matrix interactions critical for tissue organization.
- At invadopodia plasma membranes, integrin alpha-3/beta-1 provides a collagen-dependent docking site for fibroblast activation protein (FAP/seprase), facilitating adhesion, invadopodia formation, and matrix degradation to promote cell invasion.
