Biological Background: IL-2 (Interleukin-2) Function and Localization
- Interleukin-2 (IL-2), also known as T-cell growth factor (TCGF), is a secreted cytokine of the interleukin family.
- IL-2 is primarily produced by activated CD4-positive helper T-cells, with minor production by activated CD8-positive T-cells and natural killer (NK) cells. Related references: PMID:6438535
- It binds to a receptor complex composed of either the high-affinity trimeric IL-2R (IL2RA/CD25, IL2RB/CD122 and IL2RG/CD132) or the low-affinity dimeric IL-2R (IL2RB and IL2RG). Related references: PMID:16293754, PMID:16477002
- Receptor engagement triggers downstream signaling via phosphorylation of JAK1 and JAK3, leading to STAT5 activation and the stimulation of PI3K and MAPK pathways. Related references: PMID:7973659, PMID:8580378, PMID:25142963
- IL-2 functions as a T-cell growth factor and enhances the cytolytic activity of NK cells. Related references: PMID:6608729
- It promotes strong proliferation of activated B-cells and subsequent immunoglobulin production. Related references: PMID:6438535
- IL-2 plays a pivotal role in regulating the adaptive immune system by controlling the survival and proliferation of regulatory T-cells, and it participates in the differentiation and homeostasis of effector T-cell subsets, including Th1, Th2, Th17, and memory CD8-positive T-cells.
- The mature protein is a glycosylated, disulfide-bonded monomer that is secreted from cells.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the mature IL-2 sequence (amino acids 21–153) without the signal peptide, ensuring specificity for the processed, secreted form of human IL-2.
- For intracellular flow cytometry, optimize fixation and permeabilization protocols; consider using PMA and ionomycin stimulation of T-cells to enhance IL-2 production prior to staining.
- The PE conjugation allows direct detection without secondary reagents; titrate the antibody to determine the optimal concentration for your sample system.
- Cross-reactivity has only been confirmed for human IL-2; if using other species, validate specificity experimentally.
