Biological Background: IL-10RA/CD210 Function and Localization

  • Interleukin-10 receptor subunit alpha (IL-10RA), also designated CD210, CDw210a, IL-10R1, or IL10R, is the ligand-binding subunit of the IL-10 receptor complex, a class II cytokine receptor.
  • IL-10RA binds the anti-inflammatory cytokine IL-10 and, upon binding, induces a conformational change in IL-10RB, enabling the formation of a heterotetrameric receptor complex consisting of two IL-10RA and two IL-10RB subunits. Related references: PMID:16982608
  • This assembly triggers the activation of the constitutively associated kinases JAK1 (via IL-10RA) and TYK2 (via IL-10RB), leading to phosphorylation of specific tyrosine residues in the intracellular domain of IL-10RA and subsequent recruitment and activation of STAT3.
