Biological Background: IgA1 Function and Localization
- Immunoglobulin heavy constant alpha 1 (Ig alpha-1 chain C region) is the constant region of the IgA1 heavy chain, encoded by the IGHA1 gene. It is a major immunoglobulin subclass in humans, existing as both secreted and membrane-bound isoforms.
- As part of IgA antibodies, the constant region mediates humoral immunity. Membrane-bound IgA serves as the B cell receptor, triggering B cell activation upon antigen binding. Secreted IgA participates in the elimination of antigens. Related references: PMID:20176268 PMID:22158414
- The antigen binding site is formed by the variable domain of one heavy chain paired with a light chain. Diversity is generated through V(D)J rearrangement, and affinity maturation occurs via somatic hypermutation after antigen exposure. Related references: PMID:17576170 PMID:20176268
- IgA1 is the predominant immunoglobulin in body secretions such as saliva, tears, and mucosal surfaces, providing first-line immune defense. Related references: PMID:2241915
- Alternative splicing produces both secreted and membrane-bound forms of IgA1. The secreted form is found in bodily fluids, while the membrane-bound form is displayed on the surface of B lymphocytes.
- The protein is a glycoprotein with intrachain disulfide bonds and an immunoglobulin domain structure. Key associated keywords include adaptive immunity, secreted, cell membrane, and glycoprotein.
Experimental Guidance and Technical Tips
- Use this PE-conjugated antibody for direct flow cytometry detection of human IgA1 on B cell surfaces or in intracellular staining after permeabilization.
- The immunogen (amino acids 99-353 of the constant region) ensures subclass specificity; validate absence of cross-reactivity with other immunoglobulin isotypes in your sample system.
- For optimal FC results, titrate the antibody and include appropriate isotype controls and Fc blocking steps.
- Protect from light due to PE conjugation; use freshly prepared samples.
