Biological Background: EGFR Function and Localization

  • Receptor tyrosine kinase binding EGF family ligands (EGF, TGFA/TGF-alpha, AREG, epigen/EPGN, BTC/betacellulin, epiregulin/EREG and HBEGF/heparin-binding EGF) and activating several signaling cascades, including RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLCgamma-PKC and STATs modules. Related references: PMID:10805725 PMID:27153536 PMID:2790960 PMID:12297049 PMID:15611079 PMID:17909029
  • Directly phosphorylates other proteins like RGS16, activating its GTPase activity, and MUC1, increasing its interaction with SRC and CTNNB1/beta-catenin. Related references: PMID:11602604 PMID:11483589
  • Positively regulates cell migration via interaction with CCDC88A/GIV; plays a role in enhancing learning and memory performance and in mammalian pain signaling (long-lasting hypersensitivity). Related references: PMID:20462955
  • Isoform 2 may act as an antagonist of EGF action.
  • Acts as a receptor for hepatitis C virus (HCV) in hepatocytes, mediating entry by promoting CD81-CLDN1 receptor complex formation.
  • Ubiquitously expressed; isoform 2 is also expressed in ovarian cancers.
  • Subcellular localization: Cell membrane, endoplasmic reticulum membrane, Golgi apparatus membrane, nucleus membrane, endosome, endosome membrane, nucleus, secreted.

Experimental Guidance and Technical Tips

  • For flow cytometry, the PE conjugate provides high brightness and is suitable for multicolor panels. Use appropriate compensation settings.
  • The immunogen corresponds to the extracellular domain (aa 335-525), enabling detection of surface EGFR without permeabilization. For intracellular staining, permeabilization may be required.
  • Validate with appropriate negative controls (e.g., EGFR-negative cell lines or isotype control) and consider titrating the antibody to optimize the signal-to-noise ratio.
  • EGFR is a glycoprotein with multiple isoforms and post-translational modifications; the predicted molecular weight ranges from 44 to 134 kDa. This variability should be considered if the antibody is used in other applications like Western blot.
