Biological Background: CD87/PLAUR Function and Localization
- CD87 (PLAUR) is the human urokinase plasminogen activator surface receptor, also known as monocyte activation antigen Mo3. It exists in multiple isoforms due to alternative splicing.
- The receptor is GPI-anchored and recruits and activates urokinase-type plasminogen activator (uPA) at the cell surface, converting plasminogen to plasmin to initiate extracellular matrix degradation and tissue remodeling. Related references: PMID:1689240 PMID:15677461
- CD87 also binds the extracellular matrix protein vitronectin, promoting cell-matrix adhesion and indirectly modulating integrin signaling, thereby coordinating cell migration and proliferation. Related references: PMID:17548516 PMID:25168639 PMID:28849762
- Subcellularly, CD87 localizes to the cell membrane, particularly on invadopodia, and a soluble form can be secreted following GPI anchor cleavage.
- It is expressed in neurons of the rolandic area of the brain and broadly in the brain (at protein level).
- Post-translational modifications include multiple disulfide bonds, N-linked glycosylation, and a C-terminal GPI anchor, essential for stability and function.
- The predicted molecular weight is approximately 37 kDa, consistent with the glycosylated mature protein.

- The immunogen covers the extracellular domain (aa 23–303) containing the uPA and vitronectin binding regions; the antibody is likely to recognize a native epitope accessible on live cells.
- This PE-conjugated antibody is suitable for direct flow cytometry. Consider titrating on a known CD87-expressing cell line (e.g., U937 or THP-1) and include appropriate isotype controls.
- As CD87 can be shed from the cell surface, use staining buffers without detergent to preserve GPI-anchored proteins; if soluble CD87 detection is needed, employ a separate assay format.
- Validate specificity in the relevant human sample system; cross-reactivity with other species has not been reported.
