Biological Background: LILRB1 (CD85j/ILT2) Function and Localization
- LILRB1 (CD85j/ILT2) is an inhibitory receptor belonging to the leukocyte immunoglobulin-like receptor (LILR) family, containing immunoglobulin-like domains.
- It functions as a receptor for class I major histocompatibility complex (MHC) antigens, recognizing a broad spectrum including HLA-A, HLA-B, HLA-C, HLA-G, and HLA-F alleles. Related references: PMID:16455647 PMID:28636952
- Additionally recognizes the cytomegalovirus class I MHC homolog UL18, inhibiting immune responses.
- Engagement of LILRB1 on natural killer cells or T cells by class I MHC molecules protects target cells from lysis. Interaction with HLA-B or HLA-E leads to inhibition of FCER1A signaling and serotonin release, and suppresses FCGR1A-mediated phosphorylation and calcium mobilization. Related references: PMID:11907092 PMID:9285411 PMID:9842885
- Recognizes HLA-G complexed with beta-2-microglobulin (B2M) and a nonamer self-peptide, triggering secretion of growth-promoting factors by decidual NK cells. Related references: PMID:19304799 PMID:29262349
- Reprograms B cells toward an immune suppressive phenotype. Related references: PMID:24453251
- Subcellularly localized to the cell membrane and also secreted.
- Expressed in B cells, monocytes, various dendritic cell subsets, decidual macrophages, and decidual NK cells.
- Post-translational modifications include glycosylation and phosphorylation, contributing to observed molecular weights of 49–71 kDa.
Experimental Guidance and Technical Tips
- The immunogen used was a recombinant fusion protein encompassing amino acids 24–458, covering the extracellular region. For flow cytometry, this antibody recognizes a cell surface epitope; ensure appropriate cell preparation and use of live-cell staining protocols.
- Due to the expression pattern, this antibody is suitable for identifying LILRB1-positive populations in human peripheral blood mononuclear cells (PBMCs) and immune cell subsets.
- The multiple molecular weights (49–71 kDa) reflect glycosylation variants; consider this when including a total protein control.
- For optimal staining, titrate the antibody to determine the best concentration for your specific cell type and experimental conditions.
- Note that the PE conjugate is light-sensitive; handle stained samples in the dark and acquire promptly.
